Facilities

The Computer-Aided Molecular Design laboratory of Yuan-Ping Pang, Ph.D., is located on the 12th floor of the Stabile Building on the Mayo Clinic campus in Rochester, Minn. Laboratory areas include:

  • A computational chemistry unit (1,342 square feet)
  • A synthetic chemistry unit with 14 6-foot fume hoods (3,002 square feet)
  • A bioassay lab (88 square feet)

Equipment

Equipment for synthesis and characterization of compounds includes:

  • Varian 400 MHz nuclear magnetic resonance (NMR) system dedicated to Dr. Pang's lab, featuring a 1H/19F/X 5 mm autotune dual broadband probe, 1H-19F[15N-31P] 3 mm indirect broadband probe, 23 channel shims and shim supply, variable temperature module, and 100-sample autosampler
  • Thermo Nicolet Avatar 370 Fourier transform infrared spectrometer (FT-IR)
  • Jasco DIP-370 digital polarimeter
  • Molecular Devices SpectraMax Plus384 microplate reader
  • Three high-performance liquid chromatography (HPLC) systems (Beckman System Gold 125P solvent module and 166P UV detector)
  • Agilent HP 6890 gas chromatograph and Agilent HP5973 mass spectrometer
  • Liquid chromatography-mass spectrometry system (Waters 1525 binary HPLC pump, Waters 2487 dual lambda absorbance detector and Waters Micromass ZQ 2000)
  • Four Biotage SP1 flash purification systems
  • Isco CombiFlash Sg 100 gradient chromatography system
  • Isotemp cooling circulator (minus 20 degrees Celsius to 20 degrees Celsius)
  • Microcentrifuge, ovens, Kugelrohr distillation apparatus, melting point apparatus, hydrogenator, high-pressure reaction vessels, rotary evaporators, analytical balances, semi-micro balance, ozone generators, temperature controllers and freeze dryer

Equipment for solid-phase combinatorial chemistry using the radiofrequency-encoded split-and-pool technique includes:

  • AccuTag-100 system (AccuTag scanning station, Synthesis Manager software and MicroKan decapping tool)
  • Forty-eight-position optical sorting indicators
  • MultiTag-96 station drops radiofrequency tags into 8x12 arrays of MicroKans or MiniKans
  • MultiCap-96 station
  • Ninety-six-position dry resin filling station for MicroKans and MiniKans
  • Reuseable glass-coated radio frequency tags
  • MicroKans, MiniKans and MacroKans reactors
  • Genevac DD4 evaporation system

Equipment for parallel synthesis includes:

  • Argonaut Quest 210 ASW (equipped for solution- and solid-phase parallel synthesis) with automated solvent wash module and chiller interface kit
  • Five mL and 10 mL Teflon reaction vessels with 7 mm and 30 mm frits
  • JULABO FP88-MW compact ultra-low refrigerated circulator

Equipment for zebrafish experiments includes:

  • Leica M165 FC fluorescent stereo microscope with Leica IC80 HD camera, KL 1500 LCD and ErgoTube 10-50 binocular
  • Narishige three-axis course manipulator
  • Eppendorf Femtojet microinjector

Dedicated computer hardware includes:

  • Kibbutz100 (1.1 teraflops, 470 Xeon processors, 2.2/2.4 GHz, 512 MB memory/node, 8.4 TB disk)
  • SilverGene (2.7 teraflops, 590 G5 processors, 2.0/2.3 GHz, 512 MB memory/node, 4 TB Xserve RAID)
  • PowerGene (IBM SMP workstation, four POWER5 processors, 1.95 GHz, 36 MB cache, 8 GB DDR2 memory)
